Multidrug-resistant Clostridium perfringens infections are a major threat to the poultry industry. Effective alternatives to antibiotics are urgently needed to prevent these infections and limit the spread of multidrug-resistant bacteria. The aim of the study was to produce by chemical synthesis a set of enterocins of different subgroups of class II bacteriocins and to compare their spectrum of inhibitory activity, either alone or in combination, against a panel of twenty C. perfringens isolates. Enterocins A, P, SEK4 (class IIa bacteriocins), B (unsubgrouped class II bacteriocin), and L50 (class IId leaderless bacteriocin) were produced by microwave-assisted solid-phase peptide synthesis. Their antimicrobial activity was determined by agar well diffusion and microtitration methods against twenty C. perfringens isolates and against other pathogens. The FICINDEX of different combinations of the selected enterocins was calculated in order to identify combinations with synergistic effects. The results showed that synthetic analogs of L50A and L50B were the most active against C. perfringens. These peptides also showed the broadest spectrum of activity when tested against other non-clostridial indicator strains, including Listeria monocytogenes, methicillin-resistant Staphylococcus aureus, Streptococcus suis, Streptococcus pyogenes, Enterococcus cecorum, Enterococcus faecalis, as well as Gram-negative bacteria (Campylobacter coli and Pseudomonas aeruginosa), among others. The selected synthetic enterocins were combined on the basis of their different mechanisms of action, and all combinations tested showed synergy or partial synergy against C. perfringens. In conclusion, because of their high activity against C. perfringens and other pathogens, the use of synthetic enterocins alone or as a consortium can be a good alternative to the use of antibiotics in the poultry sector.

The overuse and misuse of antibiotics has contributed to the rise and spread of multidrug-resistant bacteria. To address this global public health threat, many countries have restricted the use of antibiotics as growth promoters and promoted the development of alternatives to antibiotics in human and veterinary medicine and animal farming. In food-animal production, acidifiers, bacteriophages, enzymes, phytochemicals, probiotics, prebiotics, and antimicrobial peptides have shown hallmarks as alternatives to antibiotics. This review reports the current state of these alternatives as growth-promoting factors for poultry and swine production and describes their mode of action. Recent findings on their usefulness and the factors that presently hinder their broader use in animal food production are identified by SWOT (strength, weakness, opportunity, and threat) analysis. The potential for resistance development as well as co- and cross-resistance with currently used antibiotics is also discussed. Using predetermined keywords, we searched specialized databases including Scopus, Web of Science, and Google Scholar. Antibiotic resistance cannot be stopped, but its spreading can certainly be hindered or delayed with the development of more alternatives with innovative modes of action and a wise and careful use of antimicrobials in a One Health approach.

Poly lactic acid (PLA) is a compostable, as well as recyclable, sustainable, versatile and environmentally friendly alternative, because the monomer of PLA-lactide (LA) is extracted from natural sources. PLA's techno-functional properties are fairly similar to fossil-based polymers; however, in pristine state, its brittleness and delicacy during processing pose challenges to its potential exploitation in diverse food packaging applications. PLA is, therefore, re-engineered to improve its thermal, rheological, barrier and mechanical properties through nanoparticle (NP) reinforcement. This review summarises the studies on PLA-based nanocomposites (PLA NCs) developed by reinforcing inorganic metal/metallic oxide, graphite and silica-based nanoparticles (NPs) that exhibit remarkable improvement in terms of storage modulus, tensile strength, crystallinity, glass transition temperature (Tg) value, antimicrobial property and a decrease in water vapour and oxygen permeability when compared with the pristine PLA films. This review has also discussed the regulations around the use of metal oxide-based NPs in food packaging, PLA NC biodegradability and their applications in food systems. The industrial acceptance of NCs shows highly promising perspectives for the replacement of traditional petrochemical-based polymers currently being used for food packaging.

Ginger is a commonly used spice around the world. Its bioactive compounds contain hydrophobic gingerols and hydrophilic polysaccharides. Huge physiochemical differences between these compounds and the thermal instability of gingerols impede fast and effective extraction of them using conventional methods. In this research, ionic liquid-based ultrasonic-assisted extraction (ILUAE) was applied to simultaneously extract gingerols and polysaccharides from ginger. Parameters influencing the recovery of gingerols were ionic liquid type, ionic liquid concentration, solid/liquid ratio, ultrasonic power, extraction temperature and extraction time. Compared with traditional methods, LUAE significantly increased the yield of total gingerols and shortened the extraction time. Meanwhile, ginger polysaccharides recovery reached up to 92.82% with ILUAE. Our results indicated that ILUAE has a remarkable capacity to extract gingerols and ginger polysaccharides in one step. Therefore, ILUAE represents a promising technology for simultaneous extraction of hydrophilic and hydrophobic bioactive compounds from plant materials.

Bacteria use quorum sensing (QS) systems to communicate with each other and regulate microbial group behavior, such as the secretion of virulence factors, including biofilm formation. In order to explore safe, edible agents, the potential of star anise (SA) as an anti-QS and antibiofilm agent and its possible application in milk safety were investigated. Staphylococcus aureus , Salmonella Typhimurium, Pseudomonas aeruginosa , and biosensor strain Chromobacterium violaceum were selected as test strains for QS, biofilm, and exopolysaccharide assays. The percent acidities and total plate counts were determined to evaluate the quality of biofilm-inoculated and noninoculated milk. The yield of SA extraction was 25.90% ± 0.2% (w/w). At sub-MIC, SA extract did not show any effect on bacterial growth. The production of violacein was inhibited by 89% by SA extract. The extract also inhibited the formation of biofilm by up to 87% in a dose-dependent manner. Inhibition rates of 70.45%, 42.82%, and 35.66% were found for exopolysaccharide production. The swarming motility of S. aureus was reduced by about 95.9% by SA extract. Confocal laser scanning microscopy analysis confirmed that the development of biofilm architecture was hampered. It was found that SA extract could delay the spoilage of milk. In the endeavor to avoid drug resistance, pathogenesis, and resistance to biocides while improving food safety and avoiding health hazard issues arising from synthetic chemicals, SA extract could be used as a potential QS and biofilm inhibitor.

Cell-to-cell communication or quorum sensing (QS) leads to biofilm formation and causing other virulence factors which are extreme problems for food safety, biofilm related infectious diseases etc. This study evaluated the anti-QS activity of the Amomum tsaoko extract (0.5-4 mg/ml) by using Chromobacterium violaceum a biosensor strain and biofilm formation by crystal violate assay. Experimental results demonstrated that the overall yield of Amomum tsao-ko extract was 11.33 ± 0.3% (w/w). MIC for Staphylococcus aureus (Gram positive), Salmonella Typhimurium and Pseudomonas aeruginosa (Gram negative) was 1, 2 and 2 mg/ml, respectively. A concentration of 4 mg/ml extract showed highest biofilm inhibition 51.96% on S. Typhimurium when 47.06%, 45.28% were shown by S. aureus, P. aeruginosa respectively. The damage of biofilm architecture was observed by Confocal Laser Scanning Microscopy (CLSM). A level of 44.59% inhibition of violacein production was demonstrated when the dose was 4 mg/ml. Swarming motility inhibition was observed in a dose dependent manner. Taken together, the treatment of A. tsaoko extract can deliver value to food product and medicine by controlling pathogenesis.

6-Gingerol and 6-shogaol are the main bioactive compounds in ginger. Although 6-shogaol has more and better bioactivities than its precursor 6-gingerol, the low content of 6-shogaol in ginger restricts its bioactive effects in functional foods. The traditional preparation methods of 6-shogaol are defective because of the environmental hazards and low efficiency of the processes. In this study, an efficient, easy and eco-friendly dehydration conversion of 6-gingerol to 6-shogaol is presented using an acidic ionic liquid 1-butyl-3-methylimidazolium hydrosulfate ([Bmim]HSO4) under ultrasound irradiation. The key parameters, including reaction temperature, reaction time, mass ratio of catalyst to substrate and ultrasonic power in each reaction process, were investigated. The yield of 6-shogaol reached as high as 97.16% under optimized condition. The catalyst could be separated from the reaction mixture and reused five times with only a slight loss of activity.

Polyethylene glycol (PEG) as a green solvent was employed to extract polysaccharide. The optimal conditions for PEG-based ultrasonic extraction of Dendrobium nobile Lindl. polysaccharide (JCP) were determined by response surface methodology. Under the optimal conditions: extraction temperature of 58.5°C; ultrasound power of 193W, and the concentration of polyethylene glycol-200 (PEG-200) solution of 45%, the highest JCP yield was obtained as 15.23±0.57%, which was close to the predicted yield, 15.57%. UV and FT-IR analysis revealed the general characteristic absorption peaks of both JCP with water extraction (JCPw) and PEG-200 solvent extraction (JCPp). Thermal analysis of both JCPs was performed with Thermal Gravimetric Analyzer (TGA) and Differential Scanning Calorimeter (DSC). Antioxidant activities of two polysaccharides were also compared and no significant difference in vitro was obtained.

Circadian rhythmic disorders induced by high-fat diet are associated with metabolic diseases. Resveratrol could improve metabolic disorder, but few reports focused on its effects on circadian rhythm disorders in a variety of studies. The aim of the present study was to analyze the potential effects of resveratrol on high-fat diet-induced disorders about the rhythmic expression of clock genes and clock-controlled lipid metabolism. Male C57BL/6 mice were divided into three groups: a standard diet control group (CON), a high-fat diet (HFD) group and HFD supplemented with 0.1% (w/w) resveratrol (RES). The body weight, fasting blood glucose and insulin, plasma lipids and leptin, whole body metabolic status and the expression of clock genes and clock-controlled lipogenic genes were analyzed at four different time points throughout a 24-h cycle (8:00, 14:00, 20:00, 2:00). Resveratrol, being associated with rhythmic restoration of fasting blood glucose and plasma insulin, significantly decreased the body weight in HFD mice after 11 weeks of feeding, as well as ameliorated the rhythmities of plasma leptin, lipid profiles and whole body metabolic status (respiratory exchange ratio, locomotor activity, and heat production). Meanwhile, resveratrol modified the rhythmic expression of clock genes (Clock, Bmal1 and Per2) and clock-controlled lipid metabolism related genes (Sirt1, Pparα, Srebp-1c, Acc1 and Fas). The response pattern of mRNA expression for Acc1 was similar to the plasma triglyceride. All these results indicated that resveratrol reduced lipogenesis and ultimately normalized rhythmic expression of plasma lipids, possibly via its action on clock machinery.
